Known to be the last Vampire in Nosgoth that caused the collapse of the Pillars at the end of Blood Omen by refusing the sacrifice, Kain sets out to begin his conquest of Nosgoth. Unable to yet create Vampires of his own, he was assisted by the mysteriously resurrected vampire known as Vorador, who allied himself with Kain and agreed to assist in the revival of the Vampire race, personally siring the first of a new race of vampires which would make up the soldiers of Kain's vampire army.

Umah is a vampire lieutenant of the Cabal, the vampire resistance in Meridian. She acts as both a guide and friend to the vampire lord known as Kain. Umah eventually becomes a potential love interest for Kain, yet in the end she turns traitor for Kain in favor of her position with the Cabal. Vorador took care of Umah and became her mentor. Umah saw Vorador as both her mentor and viewed him as a father figure to her from how much they respected each other. Meanwhile Vorador was the first human transformed into a vampire by Janos Audron who is one of the Ancient Vampires from Nosgoth's older history.

Ariel

Ariel's specter was bound to the Pillars of Nosgoth, in which she has haunted them for many centuries afterward. As the Guardian of Balance, Ariel is one guardian out of eight other guardians from the Circle of Nine. Her spirit remains trapped in guarding Nosgoth's Pillars since the day of their corruption. Ariel was born to serve the Pillars as Guardian of Balance at some point in the centuries of Nosgoth's early history along with the immortality gifted to her role. Ariel's magic influenced the regulation of the other magic in Nosgoth and her magical prowess was only rivaled by that of her beauty. At some point, Ariel became romantically involved with a fellow guardian known as Nupraptor, who is the Guardian of the Mind pillar. The two embarked on a controversial relationship, of which the other seven guardians may have disapproved of. Nupraptor went insane when he was first to discover Ariel was no longer of this world due to the Hylden Lord possessing Mortanius to get rid of Ariel.

Reala's signature colors are red and black. In the original Saturn game Reala had a purple ruffled collar, yellow claw-like talons for fingers and a vest that appears to be open with a diamond patterned chest. He also has a red and black-striped jester's hat that is, presumably, a part of his body and not, in fact, a hat. Reala was redesigned for "NiGHTS: Journey of Dreams"; sporting a mask called a Persona, a symbol of loyalty to Wizeman. It is gold with a pink jewel in the center with black feathers on the edges. His vest is closed over his chest and has a flame/swirl pattern. He wears red arm guards and hip-plates, which both have a flame-like design on them and gold trim. He also wears dark brown fingerless gloves and dark brown bandages on his wrists. As in the original game, his signature colors are red and black. His boots are mostly dark red, with some bright red flame patterns and a single bright red stripe interspaced in the middle of the boots. Although he keeps his red jester's hat from the original game "NiGHTS into Dreams", it now has dark red stripes instead of black stripes; it also appears to be slightly less sharply angled than the first game. Reala also wears dark brown bandages on his legs, although they are mostly concealed by his boots. He also, for some reason, has pale pink nails and a pink ruff on his neck.

The Sorrow was the "Spirit Medium Soldier" of the Cobra Unit. Although he had no actual unique combat abilities like the other Cobras, The Sorrow was gifted with the powers of a medium. He was able to summon the dead and assume their combat abilities by bringing their spirits into himself. Communicating with dead soldiers also gave him several advantages during a battle, such as learning of enemy positions. Like the other Cobras, his codename reflected the emotion he brought to the battlefield.

Shuji Shimada (嶋田 習次 , Shimada Shūji) is the first enemy encountered in "Siren: Blood Curse". He also acts as one of the game’s secondary antagonists. He is a police sergeant with a bizarre gait. The Shibito can mimic human behavior, capable of speech and are not easy to stop with doors or windows, capable of functioning as though their reanimated bodies were not possessed.

Yami Bakura appears as a recurring secondary antagonist for most of the series, until the final arc where he is the main antagonist. Fiercely loyal to his people, Vorador viewed humanity with contempt, and his exploits against the Sarafan and the Circle of Nine became legendary over several centuries.
