Trixie appears to be a huge fan of Starswirl the Bearded and dressed up as him during her Ogres and Oubliettes session. Trixie is an avid role-player and fan of Ogres and Oubliettes, and seems to fill the role of a wizard in her group. She appears to be pretty good friends with Big McIntosh and Snips and Snails, though uses the latter two has her lackies. Trixie is a skilled singer and her band, Trixie and the Illusions, was a serious contender for the winner of the Battle of the Bands, only beaten out by The Rainbooms and The Dazzlings respectively. Despite her talents as a musician, Trixie's passions lie in street magic and magician work, which she is actually not good at, at all. Most of her illusions are nothing more than basic smoke and mirrors that anyone can see through and her lack of skill makes her more complex illusions extremely dangerous for her assistants. Notably, she nearly cutting Spike in two with a dagger.Although good-hearted, Trixie is not very honest and will do anything to bask in the spotlight, even if it's through work she didn't even do. Likewise, Trixie is horrible at math and may even be insecure in her abilities, as she accused Fluttershy of cheating at math with little evidence.Trixie is extremely theatrical and a little self-absorbed. She proudly proclaims being great and powerful, but seems to be self-aware enough to realize this to an extent. She can be rude and arrogant, but not outwardly malicious to others. She enjoys eating small snacks like Peanut butter crackers and is a talented singer and musician. She's a fan of role-playing and is a member of the Ogres and Oubliettes club. As to be expected, her theatrical personality causes her to overreact there, but she appears to be good friends with other members of the club and enjoy their company.

During the first film, Sunset Shimmer is shown to be self-absorbed, condescending, manipulative, and dishonest. She frequently insults and belittles others and thinks little of it. According to human Fluttershy, Sunset makes life miserable for "anyone who stands in her way." As a result, most of the Canterlot High School student body is afraid of her. She is also not above making threats and blackmail in order to realize her goals. When Sunset puts on the element of magic, she transforms into a demonic visage of herself. While in this form, she becomes more ruthless, going so far as attempting to dispose of Twilight. After she is defeated, Sunset Shimmer becomes immediately repentant and apologizes for her actions, appearing to renounce her prior dishonest and unkind ways. In the second film, Sunset Shimmer has completely discarded her old self and starts believing in friendship, being kind toward others and befriending former enemies. With an exception of Rainbooms, the rest of the students still bearing resentment towards her and mercilessly willing to not forgive her for what she had done to everyone she hurts. She is also deeply repentant for her actions in the first film, referring to her transformed self as a "raging she-demon". However, she suffers from a lack of confidence, keeping silent in situations she feels it isn't her place to speak up and falling prey to the Dazzlings' taunting. During the final battle, she finally overcomes her insecurities and joins the Rainbooms in defeating the Dazzlings, gaining a half-pony form in the process. By the end of Rainbow Rocks, Sunset starts writing friendship reports to Princess Twilight using the book she once used to communicate with Princess Celestia.

Like her pony counterpart, Rarity is very beautiful & generous and has a keen eye for fashion. She disguises Twilight to hide her from the CHS students' ridicule, and she comes up with the idea of handing out pony ears and tails. Rarity also likes doting on the dragon-turned-puppy Spike, finding him "so adorable" and letting him sleep in her lap at the Sweet Shoppe. Rarity's generosity and love for fashion carry over into the second and third films as well. In Rainbow Rocks, she greatly concerns herself with making a good stage impression, and in Friendship Games, she says putting effort into clothes and spending time on my friends are what she lives for. Rarity also has a bit of a fascination with royalty, as she is excited to learn that Twilight Sparkle is a princess and has her own castle.

Like her pony counterpart, Fluttershy is typically very timid, shy and soft-spoken. She speaks only in barely audible whispers when she first meets Twilight in the first film, and she shrinks away from the spotlight in Rainbow Rocks. Fluttershy has a deep adoration for animals; she instantly opens up to Twilight once she sees Spike for the first time, she regularly sneaks her pets to school in her backpack, and she volunteers at an animal shelter. When Spike reveals that he can talk and is actually a dragon, Fluttershy becomes very excited to meet a talking animal. Fluttershy's strongest trait is her kindness, as displayed in her interactions with animals and various characters. In Friendship Games, she tries to cheer up human Twilight despite her being from a rivaling school. She is also more confident and self-assured in the second and third films and tie-in animated shorts than she is in the first film

Pinkie Pie shares many of her pony counterpart's character traits and cartoonish quirks, including having a very cheerful and sociable personality and performing various comedic gags. At one point in the first film, she picks one of the balloon patterns off her skirt and inflates it like an actual balloon. In the second and third films and tie-in animated shorts, Pinkie displays more cartoonish gags. She has a tendency to skip instead of walk normally, she has a refrigerator stocked with cans of whipped cream, and she tends to pop out of random places such as a locker and air vent. Though she doesn't own a party cannon in the first film, she uses her kick-drums as a party cannon in the second film, and she owns two party cannons by the third film. Above all else, Pinkie's strongest trait is her love of spreading joy to others.

Once Twilight convinces Applejack to talk to Rainbow about the misunderstanding, all five friends reconcile and help Twilight get voted for Princess of the Fall Formal.When Sunset puts on Twilight's crown and transforms into a "raging she-demon", Applejack briefly channels the element of honesty and gains pony-like attributes. Applejack reappears in the sequel My Little Pony Equestria Girls: Rainbow Rocks and its tie-in animated shorts, where she plays bass in her friends' band, the Rainbooms. She first appears helping her friends make a sign for the CHS Musical Showcase. At band rehearsal, she helps comfort Sunset Shimmer of all the bad memories of what happened at the Fall Formal. After they perform one song, she wonders why she and her friends gain pony ears and a longer ponytail while they perform even though Princess Twilight took her crown with her when she went back to Equestria.Throughout the film, Applejack frequently argues with Rarity about her concerns about the band's stage costumes, finding them to be inconsequential to the band. While trapped under the Battle of the Bands stage, Applejack and her friends reconcile after hearing Sunset Shimmer's speech, and Applejack finally decides to let Rarity use her band outfits for their performances.

Like her pony counterpart, human Twilight is shy, well-spoken and highly intelligent. She holds a great interest in knowledge and science, being able to assemble intricate scientific devices and solve complex mathematical equations, and her intellect rivals that of Sunset Shimmer. Unlike pony Twilight, however, human Twilight is socially similar to how she appears in the first episode. She is a bit of a wallflower, acting very awkward and timid around others, especially the standoffish students of Crystal Prep. Despite her problems in socializing, Twilight is eager to connect with people on a personal level, demonstrated in her interactions with Fluttershy, when the two share their love for their pets, Applejack, whom she hugged for helping her in the Tri Event Relay, and Pinkie Pie, at the Crystal Prep welcoming party. Twilight is shown increasingly distressed by the apparent harm caused by her magic-sensing and storage device to the point where Sunset's eventual accusations leave her distraught. Twilight lacks self-confidence when under extreme external (especially social) pressures. During the archery competition in the Tri-Event Relay in the Friendship Games, she is so affected by performance anxiety and Sour Sweet's anger at her poor aim that she begins to cry and her legs threaten to collapse under her. When transformed into Midnight Sparkle, Twilight becomes selfish and malicious, consumed with desire to learn everything about magic even at the cost of her entire world. However, when Spike calls out to her, sad to see what she had become, her love for him briefly brings back her sanity. When brought back to normal, she retains her normal personality and regrets what she did, but feels better when Sunset reassures that she'll be forgiven.
