Yorihime and her older sister Toyohime were tasked by Lord Tsukuyomi with carrying out Human Instrumentality Project, a plot to purify the Earth, and the Moonlight Descent Ceremony, a plan to reawaken the Moon so it can reunite with it.
Yorihime and Toyohime tracked down Satori and Koishi Komeiji, two of the last remaining satoris on Earth. Approaching Satori, Yorihime presented her with artificial bodies for her and her sister to possess while Toyohime enlisted her help in carrying out the Human Instrumentality Project, which would give everyone on Earth third eyes and ensure the Komeiji sisters wouldn't be alone. Satori reluctantly agreed, and both she and Koishi were brought to Gensokyo in preparation for the Moonlight Descent Ceremony.
The Watatsuki sisters also made a deal with Koakuma, letting her keep Patchouli to herself if she helped them in their plans. As part of the deal, Koakuma would become the one to inherit the powers of all the Gensokyo residents who died as part of the Moonlight Descent Ceremony.
During the early stages of the Moonlight Descent Ceremony, Koakuma manipulated Koishi's rampage to lure Yukari Yakumo out, while the Watatsuki sisters dispatched Reisen II to assassinate the boundary youkai.
While her sister led the Lunarian occupation of Earth in the Outside World, Yorihime went to Gensokyo to guard the Great Hakurei Border and keep anyone from destroying the gateway. She was confronted by Reimu Hakurei, Reisen Udongein Inaba, Remilia Scarlet and Kanako Yasaka when they arrived to stop her. Reisen attempted to talk her down, but Yorihime coldly sliced her head in half. Yorihime then faced off against Reimu, Remilia and Kanako and killed the latter two. Before Yorihime could kill Reimu however, the Dragon God descended and struck her right eye. Despite this, her and her sister's plans would come to fruition when Flandre Scarlet destroyed the Great Hakurei Barrier, forcing all of Gensokyo's residents into the outside world just as they
